SPECIFIC USE AND DESCRIPTION

  • The stains used in Bacteriology are used to help the morphological visualization of bacteria, which due to their insignificant size are practically transparent.
  • Although it is not its main application, the simple coloring with Safranina facilitates the observation of germs, which stand out for their pink color. It is used mainly as a contrast dye in Gram staining, replacing the diluted fuchsin that was used above, since it better differentiates Gram negatives.
  • It can also be used as in simple dyes, although it is usually preferred to other dyes that give a more pronounced hue. It is advisable to carry out the staining from young cultures (24-36 h).
  • Exclusive use as laboratory reagent Supplied with explanatory leaflet of the instructions for use. Presented in opaque amber PET bottles with a white sealed cap.

SIMPLE COLORING WITH SAFRANINE

Method:

  • Take a clean and degreased slide. (eg codes B100001, etc. general cat.).
  • Make a fine and homogeneous spread. It can be made, directly from a broth culture, from a bacterial suspension obtained by homogenizing 1-2 colonies in saline solution, or from a direct sample from a patient.
  • Let dry, and fix the extension by passing the slide over the flame of a bunsen burner. Avoid overheating.
  • Place the slide on a staining rack (eg codes S002, S003 etc. cat general).
  • Stain the smear by flooding the slide with SAFRANINE, for 2 minutes.
  • Wash with water using a washing bottle (eg codes 443000 and 191638 cat.
  • general) until the washing liquid is no longer purple.
  • Let it drain vertically (see drippers codes CP24 and CP30) and dry at room temperature.
  • Visualize with an immersion objective, placing a drop of oil between the smear and the lens.

Reading :

  • Bacteria and other elements appear pink.

Storage:

  • We recommend keeping the product between 15 and 25ºC until the expiration date indicated on the label.

Expiration:

  • Two years from the date of preparation.
